Exogenous Ascorbate Administration Elevates Testicular Oxidative Damage and Histological Injuries in Rats after Busulfan Treatment

Several anticancer drugs are coadministered with ascorbate (ASCB) to complement their cytotoxic effects. However, it is not known if the treatment regimen prevents collateral oxidative damage non-target sites. The current study evaluated effect of ASCB cotreatment on testes young adult rats treated an drug, busulfan (BUS). About 40 Wistar were arbitrarily assigned into four groups (N = 10), namely, control (<0.2% dimethyl sulfoxide vehicle), BUS (4 mg/kg b.w.; intraperitoneally for 4 days), + mg BUS/kg b.w. days 100 ASCB/kg 14 and 7 prior start injection), (100 21 days). At end study, reduced spermatogenesis score index, superoxide dismutase activity, total decreased hydrogen peroxide level, elevated catalase activity nitrite concentration much more than alone ( P < 0.05 ). Other observations included malondialdehyde DNA damage, diminished glutathione in animals. Interestingly, administration raised testicular beyond values id="M2"> ) antioxidant status testes. Histological aberrations many single layers germ cells, shrinked tubules, vacuolated structures epithelium ASCB, similar those BUS-treated In conclusion, deregulated redox caused a dramatic consumption which enhanced by exogenous resulting damage.
